I was wondering how we can enhance our English knowledge about first question which is a general question. I mean I personally do not have problem talking about simple subjects but when it comes to difficult subjects like science or something like this, I have problem. So, how can I practice this part for the exam? I very much appreciate your help in advance.

Nafis, do you mean Speaking Question 1? These are based on familiar topics. Or do you mean a different question from the Speaking test? Here is a list I found for question 1 that you can practise. I recommend recording them on your MyEC page!: http://www.english-test.net/forum/ftopic19870.html

There have been some slight adaptations to some speaking tasks on the iBT TOEFL, according to some of my students - who reported some slightly different speaking question formats on the most recent applications of the test.
